The influenza virus, also known as "the flu", is an infectious disease that has constantly affected the health of humanity. There is currently no known cure for Influenza. The Center for Innovations in Medicine at the Biodesign Institute located on campus at Arizona State University has been developing synbodies as a possible Influenza therapeutic. Specifically, at CIM, we have attempted to design these initial synbodies to target the entire Influenza virus and preliminary data leads us to believe that these synbodies target Nucleoprotein (NP). Given that the synbody targets NP, the penetration of cells via synbody should also occur. Then by Western Blot analysis we evaluated for the diminution of NP level in treated cells versus untreated cells. The focus of my honors thesis is to explore how synthetic antibodies can potentially inhibit replication of the Influenza (H1N1) A/Puerto Rico/8/34 strain so that a therapeutic can be developed. A high affinity synbody for Influenza can be utilized to test for inhibition of Influenza as shown by preliminary data. The 5-5-3819 synthetic antibody's internalization in live cells was visualized with Madin-Darby Kidney Cells under a Confocal Microscope. Then by Western Blot analysis we evaluated for the diminution of NP level in treated cells versus untreated cells. Expression of NP over 8 hours time was analyzed via Western Blot Analysis, which showed NP accumulation was retarded in synbody treated cells. The data obtained from my honors thesis and preliminary data provided suggest that the synthetic antibody penetrates live cells and targets NP. The results of my thesis presents valuable information that can be utilized by other researchers so that future experiments can be performed, eventually leading to the creation of a more effective therapeutic for influenza.

This study analyzes various reasons why some Americans find the subject of ghosts and ghost hunting so fascinating. A recent poll suggests that nearly half of Americans believe in ghosts. Thousands of ghost hunting societies exist in which members try to find proof of a supernatural world. Hundreds of films featuring paranormal elements have been released over the past few decades. The prevalence of this subject warrants further investigation regarding its appeal. Ghost stories have been passed down from one generation to the next for millennia in literature and by word of mouth. There have been accounts of ghosts haunting homes and families all over the world. Edgar Allen Poe and Nathaniel Hawthorne introduced the American horror story, which is characterized by themes that strike Americans where they care most. These stories transgressed from literature into film and television where they began reaching widespread audiences and continue to exist and thrive today. However, for as long as there have been people interested in ghost stories, there have been those interested in ghost hunting. This hobby became widely popular in the late twentieth century when popular films made it acceptable to discuss the possibility of a haunting in the home. Reality television then showed that anyone could be an investigator. Even though the topic of ghosts can be frightening and disturbing to some, others find ghost stories and ghost hunting very appealing. The purpose of this study is to understand the motivations of these investigations and the attraction to these tales. Over the course of this research, two interviews with local ghost hunters were conducted to understand their unique perspective on the subject. Further, public forums were analyzed to determine popular opinions regarding these appeals. Psychologists have proposed various theories regarding the appeal of ghosts in popular media, which provides a theoretical framework for the public opinions gathered from the forums. There is very little research regarding the motivations of ghost hunting, which is alarming. Many people have decided to dedicate their lives to this hobby, so why has so little research been conducted on the subject?

The influenza virus is the main cause of thousands of deaths each year in the United States, and far more hospitalizations. Immunization has helped in protecting people from this virus and there are a number of therapeutics which have proven effective in aiding people infected with the virus. However, these therapeutics are subject to various limitations including increased resistance, limited supply, and significant side effects. A new therapeutic is needed which addresses these problems and protects people from the influenza virus. Synbodies, synthetic antibodies, may provide a means to achieve this goal. Our group has produced a synbody, the 5-5 synbody, which has been shown to bind to and inhibit the influenza virus. The direct pull down and western blot techniques were utilized to investigate how the synbody bound to the influenza virus. Our research showed that the 5-5 synbody bound to the influenza nucleoprotein (NP) with a KD of 102.9 ± 74.48 nM. It also showed that the synbody bound strongly to influenza viral extract from two different strains of the virus, the Puerto Rico (H1N1) and Sydney (H3N2) strains. This research demonstrated that the 5-5 synbody binds with high affinity to NP, which is important because influenza NP is highly conserved between various strains of the virus and plays an important role in the replication of the viral genome. It also demonstrated that this binding is conserved between various strains of the virus, indicating that the 5-5 synbody potentially could bind many different influenza strains. This synbody may have potential as a therapeutic in the future if it is able to demonstrate similar binding in vivo.

The addition of aminoalkyl-substituted α-diimine (DI) ligands to bis(1,5 cyclooctadiene) nickel (or (COD)2Ni) resulted in the formation of two new nickel complexes with the general formula of (Me2NPrDI)2Ni and (PyEtDI)2Ni. Investigation of these complexes by 1H NMR spectroscopy revealed diimine coordination but also the absence of amine arm coordination. Using the 1H NMR spectra in conjunction with structures determined through single crystal X-ray diffraction, the electronic structure of both complexes was described as having a Ni(II) metal center that is antiferromagnetically coupled to 2 DI radical monoanions. A greater ligand field was sought by replacing the pendant amines with phosphine groups on the DI ligands. This yielded ligands with the general formula (Ph2PPrDI) and (Ph2PEtDI). Upon addition to (COD)2Ni, each ligand immediately displaced both COD ligands from the Ni0 center to produce new κ4 N,N,P,P complexes, (Ph2PPrDI)Ni and (Ph2PEtDI)Ni, as observed via single crystal X-ray diffraction and NMR spectroscopy. Reduction of the DI backbone was observed in both complexes, with both complexes being described as having a Ni(I) metal center that is antiferromagnetically coupled to a DI radical monoanion. In addition to alkylphosphine substituted DI ligands, the coordination of a pyridine diimine (PDI) ligand featuring pendant alkylphosphines was also investigated. The addition of (Ph2PPrPDI) to (COD)2Ni produced a new paramagnetic (μeff = 1.21 μB), κ4-N,N,N,P complex identified as (Ph2PPrPDI)Ni. Reduction of the PDI chelate was observed through single crystal X-ray diffraction with the electronic structure described as having a low-spin Ni(I) metal center that is weakly coupled to a PDI radical monoanion (SNi = 1/2). The ability of the three Ni complexes to mediate the hydrosilylation of several unsaturated organic substrates was subsequently investigated. Using a range of catalyst loadings, the hydrosilylation of various substituted ketones afforded a mixture of both the mono- and di-hydrosilylated products within 24 hours, while the hydrosilylation of various substituted aldehydes afforded the mono-hydrosilylated product almost exclusively within hours. (Ph2PEtDI)Ni and (Ph2PPrPDI)Ni were identified as the most effective catalysts for the hydrosilylation of aldehydes at ambient temperature using catalyst loadings of 1 mol%.

This thesis aims to address the ethics of keeping the big cats, such as lions, tigers, and leopards, in zoos. It is a practice that has generated some controversy in light of scientific studies reporting stress among wide-ranging animals in captive enclosures, as well as in the context of wider discussions in animal welfare and conservation ethics in zoos. A driving question for this project, therefore, was "What are the arguments for and against keeping large felids in zoos/captivity?" This thesis examines the historical and current ethical approaches to evaluating the ethics of maintaining big cats in zoos. Due to many of the big cat species listed as endangered species on the IUCN redlist, the species-centered approach to zoo ethics is becoming the common viewpoint, and, as a result, zoos are deemed ethical because of their contribution to ex situ conservation practices. Further, the ethical arguments against zoos are minimized when the zoos provide suitable and appropriate enclosures for their large felids. Of course, not all zoos are created equal; the ethics of zoos need to be evaluated on a case-by-case basis, but in general, it is ethical to maintain big cats in zoos.

The American English and Culture Program (AECP) at Arizona State University is an intensive language program that has taught English to speakers of other languages from over 115 countries. This study focuses on English education from five of those countries by examining the similarities and differences between AECP and English education in those countries, as well as analyzing the concerns about English education in these countries and how they may impact students who come to AECP. Those countries are Saudi Arabia, China, Japan, Korea, and Kuwait. The primary characteristics that are analyzed are history of English in relation to that country, the goals of English learning, the teaching methods used in the classes, and textbook content. The implications of this study are to help EFL educators learn about their students' backgrounds in the English language through learning the students' countries' various histories and difficulties concerning English, thus allowing them to help students better transition into the English programs such as AECP. This study also shows what research is readily available about English education in other countries, and reveals that there is a lack of research in some aspects of English education for some countries.

New-onset diabetes after kidney transplantation (NODAT) occurs in 20% of kidney transplant patients. In 5 patients who are at risk for new-onset diabetes after kidney transplantation, skeletal muscle gene expression profiling was performed both before and after kidney transplant. The differences in gene expression before and after transplant were compared in order to identify specific genes that could be linked to developing NODAT. These findings could open new avenues for future research.

The nonmedical use of prescription ADHD medications by American college students for the purpose of enhancing academic performance has been well-documented for more than a decade. A closer look at this trend through the lens of biomedicalization theory shows us that this behavior, which is often dismissed as a simple abuse of the system by deviant young people, is consistent with broader trends in post-modern medicine and health. The goal of this project was to describe the biomedicalization of studying by illustrating the historical, sociocultural, and politico-economic roots of nonmedical stimulant use by college students in the United States. An online survey was administered to students at ASU and responses from students studying in the College of Liberal Arts and Sciences (n=233) were analyzed. A 12.4% lifetime prevalence of illicit stimulant use was found among the sample population. Thematic analysis was performed on students' open-ended commentary and several themes related to the biomedicalization framework were uncovered including Necessity, Identity, Inextricability from academia, and Trust in the system. Through these themes it was found that many of the theorized transitions of biomedicalization were visible in the data set, confirming the idea that nonmedical stimulant use is embedded in the complex network of processes that represents post-modern medicine today.

In vitro measurements of cellular respiration have proven to be key biomarkers for the early onset of tumor formation in certain pathological mechanisms.1 The examination of isolated single cells has shown promise in predicting the onset of cancerous growth much earlier than current methods allow.2 Specifically, measurements of the oxygen consumption rates of precancerous cells have elucidated outliers which predict the early onset of esophageal cancer.2 Single cell profiling can fit in to current pathology studies and can serve as a step along the way, much like PCR or gel assays, in detecting biomarkers earlier than current clinical methods.3 Measurement of these single cell metabolic rates is currently limited to 25 cells per experiment. It is the aim of this project to increase throughput from 25 cells to 225 cells per experiment via the implementation of new hardware and software which fit with current methods to allow the same experimental structure. Successful implementation of such methods will allow for more rapid and efficient data collection, facilitating quantitative results and nine times the yield from the same experimental manpower and funding. This document focuses on the implementation ultra high density (UHD) hardware consisting of a pneumatic molar design, angular adjustment features and a mechanical Z-stage. These components have produced the most encouraging results thus far and are the key changes in transitioning to higher throughput experiments.
